You are working in a laboratory where you are investigating pieces of an asteroid that have been brought to Earth. You are taking every safety precaution (hazmat suit, etc.) as it is thought that the asteroid pieces contain bacteria-like microorganisms from another planet. Indeed, when you look under the microscope at the pieces of rock, you see tiny bacteria-like microorganisms. These microbes double in number every few hours when you culture them in a liquid nutrient-rich broth. i.) You determine that these microbes have surprising biochemical similarities to microbes on Earth; they have nucleic acids, proteins, lipids, and carbohydrates. However, their nucleic acids are slightly different. They have molecules similar to DNA and RNA, but both their DNA and their RNA have double helix structures, and their five-carbon sugars have a novel atom (that you call "Q") instead of oxygen in the ring structure as shown at right. Only their nucleic acids have this novel atom. Furthermore, you find that this novel atom has a radioactive isotope which you call 19Q. Their proteins seem to be composed of the same components as proteins found on Earth. You also find that this alien microbe is infected by a phage-like virus, and the phage has one molecule of RNA and one molecule of DNA inside its protein coat. You wish to determine if the DNA, the RNA, or the protein of the phage is the inherited molecule. (a) Describe how you might conduct this experiment (set up the experiment). You can assume that you have a molecular biology toolkit that works on the alien molecules similar to how that toolkit works on the molecules found on Earth. You find that the alien phage RNA-like molecule is the inherited molecule. (b) Describe results of your experiment that would be consistent with the RNA-like molecule being the molecule that is the inherited one (at least for the alien phage). ii.) You next wish to determine if the phage RNA-like molecule is dividing in a conservative or semi-conservative manner. You find that it is conservative! (a) Describe how you would conduct this experiment (use approaches similar to the ones used to understand DNA replication on Earth) and (b) result that would be consistent with conservative replication.
