Texts: Please Help!, thanks! Two plasmids, A and B. They are 3.2 kb and 3.3 kb in size, and contain vector backbones, multiple cloning sites, origins of replication (ori), but have the same ampicillin (Amp) resistance gene for selection. Plasmid A has a GFP gene inside its multiple cloning site. In this project, we need to introduce a GFP gene from plasmid A into the MCS of plasmid B using HindIII+EcoRI enzymes. After digestion and ligation, you will transform them into E. coli cells and select for Amp resistance. Due to the possibility of partial digestion, all 3 vectors could re-ligate, and the related E. coli cells will grow on Amp. Testing for GFP fluorescence will reveal cells that contain plasmids A and C. In this project, we need to design a strategy to screen for colonies containing ONLY plasmid C, separating them from plasmids A and B. For that, you have the option to introduce one additional gene into plasmid A and/or one additional gene into plasmid B (these can be the same genes or different genes). These genes cannot confer resistance to another antibiotic, such as chloramphenicol or tetracycline, etc. Expected answers (examples): Plasmid A I will introduce a gene xxx because of xxxx. The selection strategy will be: XXXXX. Plasmid B I will introduce a gene xxx because of xxxx. The selection strategy will be: XXXXX. Alternatively, in Plasmid B, I can introduce another gene xxxx. The selection strategy will be: XXXXX. EcoRI HindIII 0.7kb GFP MCS EcoRI HindIII MCS 3.2kb (backbone) Plasmid A 3.3kb (backbone) Plasmid B Amp-R Amp-R EcoRI HindIII 0.7kb GFP 3.3kb (backbone) Plasmid C Amp-R
