(3 Processes of REcombinant DNA TECHNOLOGY ombinant DNA technology involves several steps in specifle Hence such as isolation of DNA. fragmentation of DNA by fidction endonucleases, isolation of a desired DNA fragment. ston of the DNA fragment into a vector, transferring the imbinant DNA into the host, culturing the host cells in a (dium at large scale and extraction of the desired product. (us examine each of these steps in some detalls. (3.1 Isolation of the Genetic Material (DNA) (all that nucleic acid is the genetic material of all organisms hout exception. In majority of organisms this is wayribonucleic acid or DNA. In order to cut the DNA with iriction enzymes, it needs to be in pure form, free from other cro-molecules. Since the DNA is enclosed within the mbranes, we have to break the cell open to release DNA along th other macromolecules such as RNA, proteins, ysaccharides and also lipids. This can be achieved by treating (abacterial cells/plant or animal tissue with enzymes such as oogme (bacteria), cellulase (plant cells), chitinase (fungus). know that genes are located on long molecules of DNA crwined with proteins such as histones. The RNA can be removed by atment with ribonuclease whereas proteins can be removed by atment with protease. Other molecules can be removed by appropriate atments and purified DNA ultimately precipitates out after the addition chilled ethanol. Thís can be seen as collection of fine threads in the spension (Flgure 11.5).
